This is a working overview of lyophilized powder, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-04-22 and is reviewed periodically as new material appears.
Identity and purity are assessed with a small set of standard techniques. Reverse-phase high-performance liquid chromatography gives a purity estimate from peak area, usually recorded at 214 or 220 nanometers, where the peptide bond absorbs. Mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidized species. Amino acid analysis or tandem mass spectrometry sequencing can verify the sequence itself. Additional quality attributes include water content, residual trifluoroacetic acid carried over from purification, and endotoxin where the material is intended for biological work.
The compound is most often distributed as a lyophilized powder, appearing white to off-white and forming a loose cake or fluffy solid. It is hygroscopic to some degree, so brief exposure to humid air can add water weight and complicate weighing. The peptide dissolves readily in water and in neutral aqueous buffers, and aqueous solubility is generally described as high, well above the concentrations used in typical assays. Some polar organic solvents are also usable, which matters when a concentrated stock is prepared before dilution into buffer.
Storage recommendations center on keeping the dry powder cold, dry, and dark. A freezer at -20 degrees Celsius or below is conventional, and desiccant is often included to limit moisture uptake. Once dissolved, the peptide is less stable, and solutions are typically kept frozen and thawed only once. Repeated freeze-thaw cycles are a common source of losses because they promote aggregation and adsorption to container surfaces. Working aliquots are therefore prepared in advance, and glass or low-binding plastic is usually preferred over ordinary laboratory plastic.
Detection in biological samples relies on mass spectrometry, typically liquid chromatography coupled to tandem mass spectrometry after peptide extraction and enrichment. Intact peptides can also be confirmed by high-resolution mass measurement together with fragmentation data. Detection windows in urine are short because the peptide is degraded by proteases and cleared quickly, and concentrations are low. Many jurisdictions treat the compound as a prohibited substance in sport, grouped with peptide hormones and related factors, while it is not an approved therapeutic product. Identity and purity statements therefore rest on certificates of analysis, ideally issued by an independent laboratory.
Material is normally supplied as a lyophilised powder in a sealed vial. The powder is hygroscopic, so exposure to humid air leads to water uptake, caking and gradual loss of the fluffy texture that indicates a good freeze-dry. Vials are best kept sealed with desiccant, protected from light and stored cold. Letting a cold vial warm to room temperature before opening reduces condensation on the contents. Purity is normally reported from a chromatographic run, and that figure applies to the batch as tested rather than to the vial after repeated opening.

Interest in the peptide grew during the 2000s and 2010s, when studies of tendon and ligament injuries in horses reported changes in lesion size after treatment. Those reports circulated widely outside the scientific literature and shaped much of the current online discussion. Subsequent reviews noted inconsistent study design, small groups, and a shortage of independent replication. Popular descriptions often blur the line between the fragment, the complete protein, and unrelated growth factors, which complicates comparisons across sources.
TB-500 is a synthetic peptide whose sequence corresponds to a short fragment near the N-terminus of thymosin beta-4, a small protein present in most mammalian cells. The fragment is commonly cited as containing the actin-binding region of the parent molecule, which is why it appears in laboratory work on cell migration and tissue repair. Suppliers distribute it as a lyophilised powder intended for research use. Its identity is defined by amino acid sequence and by the presence of an acetyl group on the N-terminal residue.
Full-length thymosin beta-4 consists of roughly forty-three amino acids and ranks among the more abundant small proteins in the cytoplasm. The fragment is much shorter, so it cannot reproduce every function attributed to the intact molecule. In cell culture, short actin-binding motifs can interfere with filament dynamics and cell movement, but such observations come from controlled experiments rather than from whole-animal work. Whether a truncated fragment produces the same effects as the parent protein remains an open question.
Thymosin beta-4 is a 43-residue actin-binding protein found in most mammalian cell types, where it participates in cytoskeletal regulation and cell migration. TB-500 represents only a short fragment of that protein and does not include the remaining residues. Whether the isolated fragment reproduces the full range of activities reported for the intact protein remains an open question. Researchers commonly treat the two as related but distinct entities when comparing results.
Published work involving this sequence spans actin-binding assays, cell-migration studies, wound-healing models, and cardiovascular or musculoskeletal experiments. Much of the biological rationale derives from in vitro systems and animal models, and the number of controlled human studies is small. Reported outcomes vary across preparations, doses, and routes, which complicates comparison between studies. Reviews generally describe the evidence base as preliminary rather than settled. Mechanistic explanations are often proposed by analogy to the parent protein rather than demonstrated directly.
TB-500 is a synthetic seven-amino-acid peptide with the sequence LKKTETQ, corresponding to residues 17 through 23 of the protein thymosin beta-4. The N-terminus is typically acetylated in the described form, giving a monoisotopic mass near 888.5 Da and an average mass of about 889 Da. The designation TB-500 is a catalogue label rather than a formal chemical name, and the same sequence appears in the literature under several alternative abbreviations. It is handled as a research reagent rather than a pharmaceutical product.
== Medical uses == Dapagliflozin is used along with diet, exercise, and usually with other glucose-lowering medications, to improve glycaemic control in adults with type 2 diabetes. In addition, several clinical studies have demonstrated that dapagliflozin can slow the progression of kidney disease, lower the risk of kidney failure, and reduce the risk of death when used alongside standard treatments such as blood pressure medications. These benefits have been witnessed in patients with chronic kidney disease and/or heart disease, regardless of diabetes. Guidelines including the European Society of Cardiology for Heart Failure and American Heart Association consider SGLT2 inhibitors such as dapagliflozin as standard therapy for people with heart failure with reduced ejection fraction, (LVEF < 40%). This is supported by two large, randomized, controlled trials and a 2023 systematic review and meta-analysis. Evidence from multiple studies indicates that the addition of dapagliflozin and other sodium‑glucose cotransporter 2 (SGLT2) inhibitors to standard heart failure therapy can reduce the risk of worsening heart failure and hospitalisation for heart failure, regardless of the presence or absence of diabetes. A reduction in cardiovascular death is robust in heart failure with reduced ejection fraction (HFrEF), type 2 diabetes (T2DM), and chronic kidney disease (CKD), but less certain in heart failure with preserved ejection fraction (HFpEF) and acute myocardial infarction (MI).
It is unlikely that a person would overdose on nicotine through smoking alone. The US Food and Drug Administration (FDA) stated in 2013 that there are no significant safety concerns associated with the use of more than one form of over-the-counter (OTC) nicotine replacement therapy at the same time, or using OTC NRT at the same time as another nicotine-containing product, like cigarettes. The median lethal dose of nicotine in humans is unknown. Nevertheless, nicotine has a relatively high toxicity in comparison to many other alkaloids such as caffeine, which has an LD50 of 127 mg/kg when administered to mice. At sufficiently high doses, it is associated with nicotine poisoning, which, while common in children (in whom poisonous and lethal levels occur at lower doses per kilogram of body weight) rarely results in significant morbidity or death. The estimated lower dose limit for fatal outcomes is 500–1,000 mg of ingested nicotine for an adult (6.5–13 mg/kg). The initial symptoms of a nicotine overdose typically include nausea, vomiting, diarrhea, hypersalivation, abdominal pain, tachycardia (rapid heart rate), hypertension (high blood pressure), tachypnea (rapid breathing), headache, dizziness, pallor (pale skin), auditory or visual disturbances, and perspiration, followed shortly after by marked bradycardia (slow heart rate), bradypnea (slow breathing), and hypotension (low blood pressure). An increased respiratory rate (i.e., tachypnea) is one of the primary signs of nicotine poisoning.

Despite the likely increased volcanism from early plate tectonics, the Earth may have been a predominantly water world between 4.4 and 4.3 Gya. It is debated whether crust was exposed above this ocean. Immediately after the Moon-forming impact, Earth likely had little if any continental crust, a turbulent atmosphere, and a hydrosphere subject to intense ultraviolet light from a T Tauri stage Sun. It was also affected by cosmic radiation, and continued asteroid and comet impacts. The Late Heavy Bombardment hypothesis posits that a period of intense impact occurred at 4.1 to 3.8 Gya during the Hadean and early Archean eons. Originally it was thought that the Late Heavy Bombardment was a single cataclysmic impact event occurring at 3.9 Gya; this would have had the potential to sterilize Earth by volatilizing liquid oceans and blocking sunlight needed for photosynthesis, delaying the earliest possible emergence of life. More recent research questioned the intensity of the Late Heavy Bombardment and its potential for sterilisation. If it was not one giant impact but a period of raised impact rate, it would have had much less destructive power. The 3.9 Gya date arose from dating of Apollo mission sample returns collected mostly near the Imbrium Basin, biasing the age of recorded impacts. Impact modelling of the lunar surface reveals that rather than a cataclysmic event at 3.9 Gya, multiple small-scale, short-lived periods of bombardment likely occurred.

== Usefulness of NTX as a biomarker == Evaluating an individual's rate of bone turnover, termed bone remodeling, directly may be important in assessing his or her potential nonsurgical treatment response as well as evaluating his or her risk of developing complications during healing following surgical intervention. To determine an individual's rate of bone turnover, numerous biomarkers are available in the body fluids that can be correlated to this rate, and one such biomarker is NTX. However, while NTX does fluctuate in a very sensitive manner in line with bone resorption patterns, they are not very specific, in that they may vary spontaneously without physiologic intervention. For example, NTX levels may drop by 50% from day to day with no treatment, thus, making NTX levels unconvincing evidence of treatment effect. Conversely, the serum CTX biomarker, described in 2000 by Rosen, appears to be a much more effective and valuable indicator of bone resorption rate.
=== Mechanical properties === The mechanical properties of articular cartilage in load-bearing joints such as the knee and hip have been studied extensively at macro, micro, and nano-scales. These mechanical properties include the response of cartilage in frictional, compressive, shear and tensile loading. Cartilage is resilient and displays viscoelastic properties. Since cartilage has interstitial fluid that is free-moving, it makes the material difficult to test. One of the tests commonly used to overcome this obstacle is a confined compression test, which can be used in either a 'creep' or 'relaxation' mode. In creep mode, the tissue displacement is measured as a function of time under a constant load, and in relaxation mode, the force is measured as a function of time under constant displacement. During this mode, the deformation of the tissue has two main regions. In the first region, the displacement is rapid due to the initial flow of fluid out of the cartilage, and in the second region, the displacement slows down to an eventual constant equilibrium value. Under the commonly used loading conditions, the equilibrium displacement can take hours to reach. In both the creep mode and the relaxation mode of a confined compression test, a disc of cartilage is placed in an impervious, fluid-filled container and covered with a porous plate that restricts the flow of interstitial fluid to the vertical direction.
=== U.S. production ceases and resumes === The United States stopped producing bulk 238Pu with the closure of the Savannah River Site reactors in 1988. Since 1993, all of the 238Pu used in American spacecraft has been purchased from Russia. From 1992 to 1994, 10 kilograms were purchased by the US Department of Energy from Russia's Mayak Production Association. Via agreement with Minatom, the US must use plutonium for uncrewed NASA missions, and Russia must use the currency for environmental and social investment in the Chelyabinsk region, affected by long-term radioactive contamination such as the Kyshtym disaster. In total, 16.5 kilograms (36 lb) have been purchased, but Russia is no longer producing 238Pu, and their own supply is reportedly running low. In February 2013, a small amount of 238Pu was successfully produced by Oak Ridge's High Flux Isotope Reactor, and on December 22, 2015, they reported the production of 50 grams (1.8 ounces) of 238Pu. In March 2017, Ontario Power Generation (OPG) and its venture arm, Canadian Nuclear Partners, announced plans to produce 238Pu as a second source for NASA. Rods containing neptunium-237 will be fabricated by Pacific Northwest National Laboratory (PNNL) in Washington State and shipped to OPG's Darlington Nuclear Generating Station in Clarington, Ontario, Canada where they will be irradiated with neutrons inside the reactor's core to produce 238Pu.

Liu's governance of Sichuan was challenged by various natural disasters, exacerbated by war. The National Gazette reported the existence of famine in Shunqing and Jintang, as well as other areas of the province. The measures Liu took to address these crises are not known. Liu Wenhui continued his emphasis on promoting education, founding the Jianguo Middle School in Chengdu and serving as its chairman. The school reportedly had high academic standards and included stringent political training. Geographically, the wealthiest and most productive areas in central Sichuan were under Liu Wenhui's sway. This included Chengdu, but also the important salt production centers of Zilujing-Gongjing and the major river port of Yibin, as well as other agricultural and commercial facilities across the Chengdu Plain. Control of the lucrative Yunnan-Sichuan of opium, salt, and weapons in Yibin in particular was dominated by his older brother Liu Wencai, who was a key part of Liu's government. Liu funded some of his garrisons with the salt tax of Leshan, earning around 800 taels per month. These resources helped sustain Liu's armies and secure his power base.

Short transit at ambient temperature is generally tolerated, but long-term storage at room temperature is not recommended. Heat, moisture, and light all accelerate degradation. Cold, dry, dark storage is the conventional choice.
Suppliers typically quote a percentage derived from reverse-phase HPLC peak area. That figure reflects the relative amount of the main peak and does not by itself confirm identity or exclude related impurities. Mass spectrometry is commonly paired with it for confirmation.
Peptides purified by reverse-phase chromatography often carry trifluoroacetate as a counter-ion, which adds mass and can affect solubility and apparent behavior in assays. Acetate and hydrochloride forms are also offered. Knowing which form is present matters when calculating how much peptide a given weight contains.
Sealed, dry and protected from light at reduced temperature is the usual laboratory convention. Allowing a cold vial to reach room temperature before opening limits condensation. Repeated opening exposes the powder to moisture and should be minimised.
